چکیده

Chronic inflammation raises the risk of the development of colorectal cancer (CRC). Green tea catechins (GTCs), which possess anti-inflammatory properties, are known to be anti-carcinogenic in a variety of organ sites, including the colorectum. This study investigated whether (-)-epigallocatechin gallate (EGCG), a candidate chemopreventive agent and major biologically-active component of green tea, and Polyphenon E (Poly E), a mixture of GTCs, suppress inflammation-related colon carcinogenesis induced by azoxymethane (AOM) and dextran sodium sulfate (DSS). Male ICR mice aged 5 weeks were given a single intraperitoneal injection of AOM (10 mg/kg body weight), followed by 2% (w/v) DSS in drinking water for 7 days to induce colitis-related colonic tumors. They also received drinking water containing EGCG (0.01 or 0.1%) or Poly E (0.01 or 0.1%) up to week 17. At week 17, treatment with EGCG or Poly E had significantly suppressed the multiplicity and volume of colonic neoplasms as compared to the AOM/DSS group, and had resulted in a lesser degree of malignancy. In addition, treatment with EGCG or Poly E decreased the protein and mRNA expression levels of Cyclooxygenase (COX)-2 and the mRNA expression of inflammatory cytokines (TNF-α, IFN-γ, IL-6, IL-12 and IL-18) in the colonic mucosa. Our findings provide evidence that tea catechins are beneficial to the suppression of cancer development in the inflamed colon.

Structural shift of gut microbiota during chemo-preventive effects of epigallocatechin gallate on colorectal carcinogenesis in mice

AIM To investigate the effect of epigallocatechin gallate (EGCG) on structural changes of gut microbiota in colorectal carcinogenesis. METHODS An azoxymethane (AOM)/dextran sodium sulfate (DSS)-induced colitis mouse model was established. Forty-two female FVB/N mice were randomly divided into the following three groups: group 1 (10 mice, negative control) was treated with vehicle, group 2 (16...

Korean Solar Salt Ameliorates Colon Carcinogenesis in an AOM/DSS-Induced C57BL/6 Mouse Model

The effects of Korean solar salt on an azoxymethane (AOM)/dextran sodium sulfate (DSS)-induced colon cancer C57BL/6 mouse model were studied. Korean solar salt samples (SS-S, solar salt from S salt field; SS-Yb, solar salt from Yb salt field), nine-time-baked bamboo salt (BS-9x, made from SS-Yb), purified salt (PS), and SS-G (solar salt from Guérande, France) were orally administered at a conce...

A tobacco-specific carcinogen, NNK, enhances AOM/DSS-induced colon carcinogenesis in male A/J mice.

To determine whether tobacco-derived carcinogens affect colon carcinogenesis, the effects of 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) on colon carcinogenesis were examined using an azoxymethane (AOM)/dextran sulfate sodium (DSS) mouse model. NNK (10 micromol) was administered to male A/J mice by a single intraperitoneal (i.p.) injection and then AOM (10 mg/kg body weight, i.p.) was ...

Lung cancer inhibitory effect of epigallocatechin-3-gallate is dependent on its presence in a complex mixture (polyphenon E).

Green tea has been shown to exhibit cancer-preventive activities in preclinical studies. However, (-)-epigallocatechin-3-gallate (EGCG) alone was shown to be ineffective in preventing lung tumorigenesis in mice by aerosol administration. In this study, Polyphenon E and Polyphenon E without EGCG were administered by aerosol delivery to A/J mice 2 weeks after carcinogen treatment and continuing d...

Adamts18 deficiency promotes colon carcinogenesis by enhancing β-catenin and p38MAPK/ERK1/2 signaling in the mouse model of AOM/DSS-induced colitis-associated colorectal cancer

ADAMTS18 is a novel tumor suppressor and is critical to the pathology of human colorectal cancer. However, the underlying mechanism is not clear. Here we generated an Adamts18-deficient mouse strain as an in vivo model to investigate the role of ADAMTS18 in the pathogenesis of colorectal cancer. In AOM/DSS-induced colitis-associated colorectal cancer, the deficiency of Adamts18 in mice resulted...
